Dilution factor = Stock concentration ÷ Target concentration
Stock volume = Target concentration × Final volume ÷ Stock concentration
Diluent = Final volume − Stock volume

What the result means

The dilution factor describes how many total parts the final solution contains for each one part of stock.

If the calculated stock aliquot is below the reliable range of your pipette, use an intermediate dilution rather than attempting the transfer directly.

Example calculation

To make 100 µL of 20 ng/µL DNA from a 200 ng/µL stock, the factor is 200 ÷ 20 = 10×. Transfer 10 µL stock and add 90 µL diluent.

  • Use concentration values in the same units.
  • Use an intermediate dilution for impractically small aliquots.
  • Add diluent with a calibrated pipette and mix completely.

Frequently asked questions

Can I use different volume units in the two volume fields?

No. Use the same volume unit throughout so the volume ratio remains valid.

Why is a target above the stock concentration rejected?

Adding diluent cannot raise concentration; concentration requires a different process.

Should I use a serial dilution for a very large factor?

Yes. Serial steps can make tiny stock transfers more practical and reproducible.

Does the calculator account for pipetting loss?

No. It calculates nominal volumes; add a justified preparation allowance separately if needed.

What if the calculated stock volume is below my pipette range?

Prepare an intermediate dilution or increase the final volume instead of relying on an unreliable transfer.
