DNA Concentration Sample Capacity Calculator

Estimate how many complete samples can be processed from an available reagent mix for DNA assay reactions. The calculation first removes a user-defined reserve for transfer loss, converts the remaining volume to whole reactions, assigns control reactions, and groups the balance by the requested replicate count. Capacity is rounded down so the result never promises a partial sample.

Usable volume = Available volume × (1 − Reserve %)
Samples = floor((floor(Usable volume ÷ Volume per reaction) − Controls) ÷ Replicates)

What the result means

The main result is the number of samples that can each receive the full replicate allocation after controls.

Confirm whether the entered per-reaction volume is the entire reaction or only the limited reagent mix being budgeted.

Example calculation

With 1,000 µL available, a 10% reserve leaves 900 µL. At 20 µL per reaction that supports 45 reactions. After 4 controls, 41 remain; at 3 replicates per sample, capacity is 13 complete samples.

Tips for better results

  • Include all required positive and negative controls.
  • Use a reserve appropriate for the pipetting method.
  • Round capacity down, never to the nearest sample.
  • Recalculate if reaction volume changes during optimization.

Frequently asked questions

Why is sample capacity rounded down?

A partial replicate set cannot complete another sample, so only whole fully allocated samples are reported.

Are control reactions included in the sample count?

No. Controls are deducted before the remaining reactions are grouped into samples.

Does reserve percentage create extra reactions?

It reduces the volume treated as usable; it is an allowance for handling loss.

What should I enter for a single measurement per sample?

Enter one replicate per sample.

Can available volume be zero?

Yes. The calculator will report zero capacity without producing an invalid result.
