Formula

Dilution factor = (sample volume + diluent volume) ÷ sample volume; original concentration = measured concentration × dilution factor.

What the result means

A 10× dilution means one part sample in ten parts total volume. Multiply the diluted assay result by 10 to estimate the original concentration.

For multiple serial steps, multiply the dilution factors for every step before correcting the measured concentration.

Example calculation

Mixing 20 µL sample with 180 µL diluent creates 200 µL total and a 10× dilution. A measured 0.35 mg/mL corresponds to 3.5 mg/mL originally.
