Starting amount = V₀ × C₀
Recovered amount = Starting amount × recovery
Final concentration = Recovered amount ÷ V₁

What the result means

The main result is the expected concentration in the resuspended sample. It includes recovery loss, so it is more realistic than simply dividing the starting volume by the final volume.

This is a mass-balance estimate. Actual recovery depends on pellet formation, aspiration loss, adsorption, solubility, and resuspension completeness.

Example calculation

For 50 mL at 2 mg/mL, the starting amount is 100 mg. At 85% recovery, 85 mg remains. Resuspending in 5 mL gives 17 mg/mL, an 8.5× concentration factor, with 15 mg estimated loss.

Tips for better results

  • Measure concentration before and after processing when recovery matters.
  • Avoid aspirating close to a loose or invisible pellet.
  • Use the smallest resuspension volume that still permits complete mixing.
  • Report both concentration and recovery; a high concentration can conceal a poor yield.
  • Keep concentration units identical before entering values.

Frequently asked questions

How is centrifuge concentration different from the volume reduction ratio?

The volume ratio ignores recovery loss. This calculator applies the recovered fraction before dividing by final volume.

Can recovery be greater than 100 percent?

No. Values above 100% usually indicate measurement variability, unit mismatch, or an incorrect blank.

What if I do not know the recovery percentage?

Use 100% for a theoretical upper bound, then compare it with a conservative scenario such as 70% or 80%.

Does pellet wetness affect the result?

Yes. Residual supernatant increases the true final volume and can lower concentration relative to the estimate.

Can I use cells per milliliter instead of mg per milliliter?

Yes, if both starting and final concentration are expressed in the same count-per-volume unit.
